Talk to any travel agency in Madurai or Rameswaram if you are looking for a private cab. Bus is also available with a good frequency. It takes around 2.5hrs by road.

Regular govt buses are available from Madurai to Rameshwaram. If you are not comfortable with them, you can take a cab

Always take a car if you are going in a group.

If you are taking a room, try to put it somewhat near the temple to avoid going back and forth since you need to do it after dipping into the sea and getting changed, and then only you can visit the temple.
